Genetic diversity analysis with rapd-pcr method at isatis floribunda boiss. ex bornm. (Brassicaceae)

Turkey is one of the richest bio-geographic regions in the world in terms of plant biodiversity, plant genetic resources and endemism. Isatis genus is distributed within the borders of our country with its 32 species and 47 taxa of which 22 are endemic. In the literature reviews, it has been observed that a molecular genetic study showing genetic diversity in the Isatis floribunda Boiss. ex Bornm. (deliızgın) has not been performed yet. The purpose of this thesis is to create RAPD markers in Isatis floribunda by using RAPD-PCR method which enables detection of genetic diversity with sensivity and efficiency and to bring molecular genetic analysis results into literature. As a result of PCR reactions, a total of 114 bands were observed. 95 of these bands were evaluated as polymorphic and 19 of them monomorphic bands. Total polymorphism rate was detected as 83.3%. Of the 15 primers used, only OPA-14 desired polymorphic RAPD products could not be amplified. Six of the remaining 14 primers (MM, OPA-08, OPA-12, OPA-13, OPA-15 ve OPA-18) achieved 100% polymorphism while eight primers had polymorphism values ranging from 25-90.9%. 31 of the 95 polymorphic bands are population spesific. This result is an important finding in terms of showing that the endemic species of Isatis floribunda which is located in Aksaray, can maintain genetic diversity within its own gene center boundaries and that the level of genetic variation among population is high. This thesis is the first scientific research on this subject; it will provide useful literature information that can be used in subsequent molecular genetic studies on Isatis floribunda species.
