Abstract (EN)

The Kalanchoe genus belongs to the Crassulaceae family and is represented by about 130 species. The most well-known species in the world is Kalanchoe blossfeldiana Poelln. ( Flaming Katy). K.blossfeldiana Poelln. are perennial plants with succulent character. It is one of the most used ornamental plants in interior designs, adding vitality to the place where it is placed due to its decorative leaves, long-life and colorful flowers. In this study, micropropagation of Kalanchoe blossfeldiana was studied. In the research, firstly, adventitious shoot regeneration was tried to be obtained by using leaf explants. For this purpose, the effects of different growth regulators and their combinations were investigated. Although the type and dose of growth regulators used vary according to the trials, in general, NAA (0.1, 0.5, 1.0 mg/L) from auxin group hormones, BAP (2.0 mg/L) and TDZ (0.1, 0.2, 0.5, 1.0) from cytokinin group hormones. mg/L) was added to the MS nutrient medium. Adventitious shoot formation rates were determined in MS nutrient media containing growth regulators. In vitro shoots were matured in a combination of ½ MS + 0.2 mg/L GA3 nutrient medium. The shoots with sufficient growth were rooted in 100% both in still water culture and in hormone-free ½ MS, MS, and MS medium with 0.1 mg/L IBA added. Rooted plantlets were successfully acclimatized in a mixture of perlite + peat (1:1), vermiculite, and a commercial seedling-growing soil. The plants were adapted to the external conditions and were taken into pots and developed until the flowering stage. It was determined that they were in the same structure as the donor plant, and no somaclonal variation was observed.
