Abstract (EN)
Cancer is a common severe disease; many people suffer from it, and it shows a high mortality rate across the world. Traditional cancer treatments including chemotherapy, surgery, and radiotherapy have some significant drawbacks such as deprivation of targeting, complications from surgery. Photodynamic therapy (PDT) is a promising method for establishing novel cancer treatment modalities as it offers a selective targeting mechanism and uses nonionizing radiation. PDT requires nontoxic light-sensitive photochemical agents, which are called photosensitizers (PS), and they can be activated with light exposure, generating reactive oxygen species (ROS), which triggers cell death. However, PSs may have some limitations, such as low water solubility, aggregation, and bioavailability, which leads to insufficient accumulation of the therapeutic target. Therefore, nanoparticle-based drug delivery systems can enhance the targeting of such agents and release them with controllable photoactivity at predetermined locations, thus obtaining desired therapeutic effects with minimal adverse effects. In this study, the PDT potential of Zinc Phthalocyanine (ZnPc) incorporated Mesoporous silica nanoparticles (MSNs) was investigated on A-549 lung adenocarcinoma cell line by analyzing dark cytotoxicity, stability, cellular uptake, and light exposure assays. As a result, ZnPc-MSN demonstrated low dark toxicity, significant cellular uptake, and potent singlet oxygen generation upon the light exposure, demonstrating their potential as an effective PS. Further research will evaluate the surface modification of ZnPc-MSN to increase cellular uptake and augment the endosomal escape efficacy of in vivo experiments. Additionally, dual PS loading will be evaluated to increase the PDT effect.
