Identification of gene flow through microsatellite DNA analysis for black sea trout(Salmo trutta Labraxpallas,1811) and brook trout (Salvelinus Fontinalismitchiil, 1814) in their hybrids (tiger trout)

Abstract (EN)

In this study, Tiger trout' (Black Sea trout Female♀×Brook troutMale♂)gene transfer was examined through microsatellite DNA analysis. ForBlack Sea trout,Brook troutandTiger trout, on the point of being 94 individuals in total; 8 spawners and 86 larvae, and 13 microsatellite loci were used. According to the analysis results, 71 different alleles were identified. In addition to identifying the average number of alleles as 5,462, the number of alleles pertaining to 13 different microsatellite loci varies between 3 and 9, and respectively T3-13 (9), SSSP2201 (9) and SSSA410 (3), STR60 (3), STR12 (3) were identified as from highest to lowest polymorphism indicative loci. Expected and observed values of spawners' larvae genotypes with frequency values vary between 0, 52-1 for DD1 X DE1, 0-1 for DD2 X DE3, 0-1 for DD3 X DE4. For our hybrid group DD1 KE1, highest value belongs to STR60 locus. For DD2 X KE4 group values vary between 0-1 and highest value was observed in BS131, STR58, SSA410, OMY7, SSA85, STR73, STR12 loci and for DD3 X KE4, values vary between 0-1. The highest values were observed in BS131, SSA85 and STR12 loci. Distinctness in gene transfer was identified between DD1 X KE1 larvae and DD2xKE4 and DD3xKE4 larvae. 41.6% of gene transfer from father and the same from mother for DD1xKE1, 50% from mother for DD2xKE4, 66.7% from mother for DD3xKE4 were was observed. However, looking at the all loci and groups, observations indicates that 52.78% of gene transfer comes from mother, 16.67% from father and 30.56% from mother+father.
