Investigation of secondary metabolites and bioactivities of Lactobacillus plantarum strain at molecular level

Abstract (EN)

In this study, the cytotoxic effects of the probiotic Lactobacillus plantarum secondary metabolite on the human breast cancer cell line were identified as MCF-7 cell line and the Drosophila melanogaster fruit fly, investigated. The IC50 value of the secondary metabolite in the MCF-7 cell line was 0.0013 gr/ml. Drosophila melanogaster doses were determined during the study and the probit analysis resultant LC50 and LC99 values were 0.24 mg/ml and 0.56 mg/ml, respectively. After application, MCF-7 cell line; RRAS-2, TP53, BCL-2, APAF-1, CASP-3, FADD, CASP-7, BOK genes and expression levels of RAS64B P53, BUFFY, DARK, DECAY, FADD, DRICE and DEBCL genes in Drosophila melanogaster were determined by RT-PCR expression of the BCL-2 and BUFFY anti-apoptotic pathway genes, in the human and the fruit fly, has been reduced. At the same time, expression increase of DECAY, FADD, RAS64B apoptotic genes was detected in Drosophila melanogaster. However, analysis of Lactobacillus plantarum secondary metabolite was performed by GC-MS method. Molecular docking was used to link the matter to the related genes. The substance detected in the secondary metabolite content and encoded as L13 has been observed to have high binding affinity in the studied genes.
