Abstract (EN)
Growth characteristics of the shoot-tip explants obtained from Lavandula angustifolia, an aromatic and medicinal plant, on various media were examined, the essential oil yields of the seedlings grown in vitro were determined and finally their phenolic constituents were analyzed. The most suitable media for germination, shoot formation and development were individually investigated. In order to observe the micropropagation potential of the plant studied, Murashige ve Skoog(MS; 1962) basal medium was supplemented with various concentrations of plant growth regulators; e.g. benzylaminopurine (BAP), 6-furfurylaminopürine (kinetin). The best germination (65 %) was achieved on MS medium supplemented with naphtalene acetic acid (NAA, 0,5 mg/L). The shoot tip explants obtained from the newly germinated seedlings were then transferred to sixteen different MS media where their growth parameters were individually examined. The best multiple shoot development was observed on MS basal media supplemented with BAP (1 mg/L) plus 0,05 mg/L NAA (0.05 mg/L). Amongst the all cultured explants, the tallest seedlings were obtained from the media containing Kinetin (1.5 mg/L) + NAA (0,05 mg/L). Shoot node explants formed callus at higher frequencies when they were cultured on a MS basal medium supplemented with l BAP (2 mg/L) + NAA (0,05 mg/ L). The extracts obtained from the shoot culture lines were analyzed qualitatively and quantitatively in terms of their four different phenolic constituents, namely ferulic acid, gallic acid, chlorogenic acid and rosmarinic acid by using HPLC. The highest essential oil yield (3.37 %, v/w) was obtained from the seedlings cultured on MS medium with kinetin 1 mg/L plus NAA (0.05 mg/L).
