Abstract (EN)

Biotechnology is currently the most popular scientific area open to innovations. Molecular studies have gained considerable importance in microbial biotechnology studies. Morphological, biochemical and molecular methods are used for the classification of microorganisms. Microorganisms were isolated and purified from deteriorated lettuce plants with industrial importance obtained from local markets. For identification, fatty acid methyl ester profiles of bacterial isolates were determined. The MIS method was used for identification in accordance with these profiles. Molecular methods, one of the rapidly developing diagnostic methods is the most accırate method used in the identification of bacteria. Amplification and sequencing of this gene using universal primers that allow the amplification of the bacterial 16S rRNA gene region has begun to be used as a standard method in phylogenetic studies. Sequence results were analyzed using BLAST to carry out identification. As a result of the sequence analysis, Bacillus pumilus, Bacillus suptilis, Bacillus thuringiensis, Bacillus cereus, Chryseo bacterium indoltheticum, Bacillus amyloliquefaciens, Pectobacteriume carotovorum, Bacillus laevolacticus and Bacillus safensis species were identified. Phylogenetic tree was formed by these species and linkage relations were examined. In the last step, destructive effects were determined based on the chitinase and phytase activity of these bacteria. This study involves the identification of the disruptive organisms which is the first step of the experiment to support the good agricultural practices of the lettuce plant with economic importance, isolation of these microorganisms and the molecular analysis of the identifications. The presence of some extracellular enzymes (phytase and chitinase) in the identified microorganisms has been qualitatively examined.
