Abstract (EN)

In recent years, scientists have become increasingly interested in compounds that are naturally present in the structure of plants and secreted as secondary metabolites in plants. In addition to the daily use of substances obtained from these compounds, it is working to use them therapeutically in the treatment of diseases such as cancer. One of these compounds, oleanolic acid, is a naturally occurring pentacyclic triterpenoid bound to betulinic acid. It has been revealed in studies that Triterpenoid compounds have the potential for farmocological action. Oleanolic acid is also known for its anti-cancer, antioxidant, antifungal, antimicrobial and antiflamatory effects. The aim of the study was to determine the cytotoxic effects of oleanolic acid on the human breast cell line and to determine the lethal effect of oleanolic acid on the larvae of Drosophila melanogaster, a good model organism. In the study, solutions at concentrations of oleanolic acid 1, 5, 10 and 20 mM were found to Be Wild Type D.melanogaster III. It has been applied to instar larvae. 10 females and 10 males were incubated 24 hours at 25 °C and 60% humidity in fresh media. Substances found in concentrations were applied directly to larvae death rates LC 50 and LC 99 values were determined as 10.4 mM and 32.5 mM. CG15530, AKT-1, Dr-PN gene expression levels of oleanolic acid on D. melanogaster were significantly determined at all concentrations applied according to the control group. But no significance was observed at the level of expression of the DR-Buffy gene compared to the control group. In MCF-7 cell lines, significance was determined in the H-prune and BCL-2 genes in the bad, BCL-2, h-prune and AKT-1 gene expression level results.
