Abstract (EN)

Safinamide is a drug active substance used in the treatment of Parkinson's disease by selectively and reversibly inhibit the MAO-B enzyme. In this thesis, the first synthesis of α-aminoamide derivatives having safinamide skeleton was aimed. For this purpose, benzyloxy benzene derivatives 4-(benzyloxy)-3-methoxybenzaldehyde (83) and 2-(benzyloxy)-4-methoxybenzaldehyde (85) were obtained from the reaction of benzyl bromide with vanillin (82) or 2-hydroxy-4-methoxy benzaldehyde (84). Amino acid methyl ester substituted benzyloxy benzene derivatives 90-95 were synthesized by reducing the imine derivatives formed as a result of the reaction of the related benzyloxy benzaldehyde derivatives with methionine, phenylalanine and tyrosine methyl esters. As a result of the reaction of compounds 90-95 with excess NH3, new analogues of Safinamide drug 76-81 were obtained with good yields.
