Molecular analysis of LAP3, MASP2 and SIRT1 genes associated with mastitis resistance in Şavak Akkaraman sheep

Abstract (EN)

Sheepare ruminant mammals used as livestock form eat, milk, and wool production in agricultural industries around the world.Genetican genomic in formation can be used to improvem any production traits, including growth rate, meat, milk, wool yield, and disease resistance. However, importantin formation abouth the functions of genes in the sheep genome is stil lacking. Mastitis is one of the most common diseases in dairy sheep, causing huge economic losses and affecting animal welfare. The disease persists in all dairy sheep production systems despite the application of hygieni corother methods. Genetic approachesare thought to provide important contribution stotheb reeding of health-related traits in farm animals. In this respect, in this thesis, LAP3-intron-12, LAP3-exon-13, MASP2-exon-11, MASP2-exon-2&3,and SIRT1-promoter gene regions polymorphism swere determined by Savak Akkaraman sheep breed. Genomic DNA was obtained from blood samples of 150 sheep of the Savak Akkaraman sheep breed. All samples were genotyped fortarget gene regions by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism method using TasI, HindII, NmuCI, PstI, TaqI, HhaI, BstUI, BsrDI, BsrI, BsaJI, VspI, SmaI enzymes.The LAP3-intron-12 region exhibited the highest heterozygosity and revealed a balanced distribution of all eles (C and T) with moderate genetic diversity. LAP3-exon-13 showed complete homozygosity, indicating no genetic diversity. MASP2-exon-11 showed high genetic diversity with the AA genotype being dominant. MASP2-exon-2&3 showed moderate genetic diversity with them ajority having the TT genotype. SIRT1-promoter exhibited moderate heterozygosity,and a C>G SNP locus revealed polymorphic patterns. Hardy-Weinberg Equilibrium analysis showed deviations for LAP3-intron-12 and MASP2-exon-11, suggesting potenti a leffects of non-random mating or genetic drift. Correlation analyses between genotypes and milkyield in Savak Akkaraman ewes revealed significant positive relation ships forth e LAP3-intron-12 CC genotype and higher milk production, while the MASP2-exon-11 BB genotype showed a significant negative correlation with lower milk yield. The sefindings provide in sights into the genetic basis of milk production traits in Savak Akkaraman ewes
