Investigation of the effect of ferulic acid against neurotoxicity induced by cisplatin in rats

Abstract (EN)
This thesis study was designed to demonstrate the possible beneficial effects of Ferulic acid, which has a large number of biological activities in alleviating experimentally generated Cisplatin-derived acute brain tissue damage. Thirty male rats were used in our study. Experiments were randomly divided into 5 groups as 6 rats in each group and groups formed as Control, Cisplatin, Ethanol, Ferulic acid and Ferulic acid+Cisplatin groups. Any drug administration was made in the Control group. A single dose of 10 mg / kg cisplatin was given as intraperitoneal and 24-hour cisplatin -induction was provided. In the Ferulic acid group, 100 mg / kg Ferulic acid was administered intraperitoneally for 5 days. Ferulic acid and Cisplatin drug applications were done as combination in Ferulic acid+Cisplatin group. On the last day of experiment, all experiments were sacrificed by high dose anesthesia and brain tissues taken rapidly. Superoxide dismutase (SOD) activity and glutathione (GSH) and malondialdehyde (MDA) levels were evaluated in all tissue samples. According to our biochemical results, in brain tissue samples, MDA levels increased in the Cisplatin group, but these levels decreased in the Ferulic acid+Cisplatin treatment group. Furthermore, SOD activity was significantly reduced in the Cisplatin group; whereas in the Ferulic acid+Cisplatin treatment group it was evaluated to be increased. Also, when the Control and Ferulic acid groups compared with Cisplatin group, we were observed that GSH levels reduced in rat brain tissue. When our biochemical results are evaluated, we can express that Ferulic acid may be a protective agent against oxidative brain tissue damage caused by cisplatin.
