Effect of Rutin on Chemerin and eNOS expressions in an experimental hypertension model in rats

Abstract (EN)

This study aimed to investigate the effect of Rutin on Chemerin (Chemerin chemokine-receptor 1-CMKLR1) and endothelial nitric oxide synthase (eNOS) expressions in the experimental hypertension model created with NG-nitro-L-arginine methyl ester (L-NAME). In the study, 28 adult Spraque-Dawley male rats were selected, 7 in each group, and divided into a total of 4 groups. Control group; 0.5 ml of olive oil was given daily by oral gavage for 21 days. L-NAME group; L-NAME was given by oral gavage together with olive oil at a daily dose of 50 mg/kg/BW for 21 days. L-NAME+Rutin group; L-NAME was given together with olive oil at a daily dose of 50 mg/kg/BW for 21 days, and subsequently Rutin at a dose of 100 mg/kg/BW was suspended in distilled water and given by oral gavage. Rutin group; Rutin at a daily dose of 100 mg/kg/BW was suspended in distilled water and administered by oral gavage for 21 days. A significant difference was determined between the groups in terms of blood pressure values of all rats measured on the 0th, 7th, 14th and 21st days of the study (p<0.05). It was determined that there was a significant increase (p<0.05) in the systolic blood pressure (SBP) values of the L-NAME group rats compared to the control group. It was determined that there was a significant decrease in MBP values in the L-NAME+Rutin group compared to the L-NAME group (p<0.05). While significant histopathological changes were detected in the kidney and aorta tissues of L-NAME group rats compared to the control group, these changes were found to be attenuated in the L-NAME+Rutin group compared to the L-NAME group. While increased CMKLR1 and decreased eNOS immunoreactivities were detected in the kidney and aorta tissues of L-NAME group rats compared to the control group (p<0.05), decreased CMKLR1 and increased eNOS immunoreactivities were noted in the L-NAME+Rutin group compared to the L-NAME group (p<0.05). As a result, it was concluded that Rutin had regulatory effects on Chemerin and eNOS immunoreactivities in the experimental hypertension model.
