Abstract (EN)

The purpose of this study is to synthesize Protoporphyrin IX, Mg2+ in the laboratory conditions of the resultant Magnesium Porphyrin which is the first step of chlorophyll synthesis in the catalysis of magnesium chelatase enzyme. Accordingly, enzymes of magnesium chelatase have been extracted from the green plant leaves and purification procedures have been applied. Dye affinity chromatography has been used to purify the magnesium chelatase enzyme and cibacron blue has used as a dye. The characterization of the synthesized dye affinity column has performed by using parameters such as infrared spectroscopy (IR), swelling tests and scanning electron microscopy (SEM) images. Subsequently, a protein mixture containing poly (HEMA-co-magnesium chelatase) column and magnesium chelatase enzyme according to the Aminoacid (monomer) Decorated and Light Underpinning Conjugation Approach (ANADOLUCA) method has immobilized to provide support material for Magnesium Porphyrin synthesis. Swelling tests have performed for the characterization of the synthesized column material, IR and SEM images have been taken. Magnesium Porphyrin synthesized by passing Protoporphyrin IX and Mg2+ solution through the column has been determined by the fluorimetric method.
