Comparison With Conventional Methods In Molecular Methods In The Sepsis Diagnosis Sepsis is a multisystem disease, typically leading to hemodynamic changes, shock, organ dysfunction and organ failure and can be life threatening.Despite all medical advances, sepsis is still a clinical entity associated with high mortality.Early diagnosis and treatment is the most important factor in determining prognosis in a positive way. The most widely used test to identify the etiology is blood culture; but efficiency is not at the desired level.Molecular tests are the future applications in this regard.In this study, the efficacy of the molecular polymerase chain reaction was investigated in comparison to blood culture for to identify the causative microorganism of sepsis. 116 patients diagnosed as sepsis were included in the study and a total number of 232 blood samples were examined. 74 of the patients were male(64%) and the other 42 were female(36%).Age of the patients ranged between 17 to 90 ( 60.74 ± 16,26).While the ratio of blood culture positivity was 47.4%, there was no positive results with PCR. A total of 232 blood cultures were collected and positivity was detected in 90 (38.79%) bottles. Gram positive isolates were the most isolated microorganisms followed by Gram negative bacteria and Candida spp. The most commonly isolated microorganisms were coagulase negative staphylococci (43.3%), E.coli (10%) and Candida spp. (10%). The negative results of PCR detected in the study is thought to be the the result of the used PCR method. The low DNA levels in samples is the probabl reason for the negative results of classical PCR. Nested PCR or multiplex real time PCR methods which may detect small fragments of 16S rRNA would be more suitable for molecular detection of microorgansims.
