Abstract (EN)
SUMMARY Propagation of European Aspen (Populus tremula L.) and Caucasian Linden (Tilia rubra DC.) Using By Tissue Culture Techniques In this study, in european aspen was studied the effect of different sucrose, BAP, BAP + IBA, kinetin, kinetin + IBA concentrations and combinations on shoot formation. For that reason, bud explants taken during the dormancy period were cultured into two medium known as the MS and WPM. It was found that the BAP into the WPM media of 0.2 mg/l BAP concentration was more effective than that of other BAP concentrations, in respect to shoot lenght and shoot and nod number. Shoot formation was not observed in BAP + IBA, kinetin, kinetin + IBA concentrations and combinations. Shoot formation was not also observed in all MS medium. The bud explants belonging the six different ages individuals were cultured into WPM of 0.2 and 0.5 mg/l BAP. The shoot and nod number and shoot length were different depending on individuals. Two different methods consisting of in vitro and in vivo were used for rooting of microcuttings and the rooting in vivo was found to be more successful for rooting and survival than in vitro In Caucasian linden, the effect of different sucrose, BAP and IBA concentration on embryo development and plantlet formation and the seeds collected in different time and cultured in sucrose media on the development of embryos were investigated. Experiments showed that when sucrose (10, 20, 30 and 40 g/l) only supplemented into both MS and WPM media, embryos grew plantlets. It was found that the most effective concentration of sucrose was 30 g/l. Embryos cultured in IBA (0.1 and 0.2 mg/l) medium became grenish and others cultured in BAP (0.1 and 0.2 mg/l) became brownish. The development of embryos of collected in different time the seeds was also different. The embryos of mature seeds were become plantlets while others collected from the immature seeds were not.
