The effects of resveratrol over antioxiant metabolism on TNBS-induced experimental colitis

Abstract (EN)

In this study, the effects of prophylactic resveratrol on TNBS induced colitis wereinvestigated, which has anti-inflammatory and antioxidant effects. Thirty-five WistarAlbinomale rats were divided into 5 groups: Drug Control, Sham Control, TNBS,TNBS+DMSO (Vehicle Control) and TNBS+RSV (n=7). Before induction of colitis,animals in vehicle and treatment groups were treated intraperitoneally by Dimethyl sulfate(DMSO) and Resveratrol (RSV) (10 mg/kg/day) respectively.After fasting the animals overnight and emptying the colons on the morning ofexperiment, inflammation was induced in the colon by the intrarectal administration of 0,8ml of a 25-mg TNBS solution dissolved in 37% ethanol in saline using an 8-cm-longcannula under ether anesthesia. Animals were sacrifised by toxic dose ether 24 hoursfollowing induction of colitis. After decapitation, the last 10 cm of the colon was excised,opened longitudinally. Longitudinal colon segments were subjected to biochemical andhistopathological examination. Statistical analyses of data were carried out using the SPSSprogram.MDA levels of TNBS and TNBS+DMSO groups were higher than Drug and Shamcontrol groups (p<0.01). RSV treatment decreased MDA levels compared to TNBS group(p<0.01). GSH-Px activity in treatment group were higher than all other groups (p<0.001).CAT activity in treatment group was also higher compared to TNBS and TNBS+DMSOgroups. However, this increase in CAT was not significant (p>0.05). While MPO activitiesof TNBS and TNBS+DMSO groups were higher than Drug and Sham Controls, there wasno decrease in MPO activity in TNBS+RSV group by RSV treatment. In histologicalexamination, tissue injury and necrosis was significantly decreased in the treatment groupcompared to TNBS colitis and vehicle group (p<0.05).In Conclusion, it has been observed that prophylactic RSV has antioxidant activityon TNBS colitis. However, failure of anti-inflammatory effects through MPO activitycould be due to that RSV dose is lower or that post treatment way is much effective thanpretreatment way.Keywords: Resveratrol, TNBS, Colitis, antioxidant, anti-inflammatory.
