Abstract (EN)
OBJECTİVE: In study, we aimed to reveal effectiveness of platelet rich plasma on degenerated chondrocyte cells by using different doses, duration and activation method of thrombocytes. MATERIALS AND METHOD: Human chondrocyte cells were used. Experimental model of chondrocyte cells were yielded. Platelet rich plasma was obtained from voluntaries with afarez method and was set 4,8x106-2.4x106- 1,2x106-6x105-3x105 doses. The first group was left intact, the second was exploded seven minutes by applying ultrasonic waves in water, the third was activated with calcium chloride and last was the control. The linear damage was created with ten microliters pipette. Cell migration was photographed with inverted microscope at magnification of x10 and at 0-4-8-24-48 hours. The pictures were evaluated by imageJ programme to describe the space between migrated cells. RESULTS: At 4-8-24. hours exploded and intact groups at all doses were found a non-significant to the control, while a significant improvement at 48 hour was observed. A calcium-activated group was not found statistically meaningfull with all doses at 4-8-24 hours but significant improvement were observed at 4,8x106- 2.4x106-1,2x106 doses at 48 hours. At 48 hour at 3x105-6x105 doses, intact and exploded groups were significantly different to control. But all groups showed significant improvement at 1,2x106 and higher doses. Exploded and intact groups showed significant improvement, at 3x105-6x105 doses at 48 hours to third group. CONCLUSION: It has been shown that all doses had pozitive effect on healing. Meanwhile the time was critical parameter of healing and the significant healing was accomplished after 48 hours.
