Varicocele, commonly seen in couples presenting to the clinic because of infertility, is the varicocele that causes the backflow of dirty blood from the testis as a result of abnormal expansion of the testicular vein. Excessive oxidative stress caused by varicocele etiology disrupts cellular homeostasis, causing fragmentation and maturation of sperm DNA, abnormal chromatin packaging and mitochondrial damage, resulting in male infertility. The aim of this study was to investigate the effects of oxidative stress-related mitochondrial membrane potential, DNA fragmentation, chromatin condensation and sperm maturation in spermatozoa in varicocele and normal cases. In this study; Biruni University Hospital Urology outpatient clinic varicocele patients (n: 24) and normal patient groups (n: 24) according to WHO criteria standard standard semen analysis was applied, followed by aniline blue (blue) staining method with sperm maturation defect, Acridine orange (orange) staining method sperm DNA fragmentation damage, sperm chromatin condensation with Chromomycin A3 staining method and the effect of oxidative stress on mitochondrial membrane potential with Nitroblue tetrazolium staining were evaluated. The evaluation of the data between the two groups was compared with student-t test analysis. According to this thesis, varicocele patients were found statistically different in terms of sperm concentration, motility and morphology in varicocele and normal patient groups (p <0.05). There was a significant difference between two groups in terms of DNA fragmentation, sperm maturation, chromatin condensation and mitochondrial membrane potential, and the effect of varicocele induced oxidative stress on mitochondrial membrane potential was determined (p <0.05). Varicocele patient groups were found to be higher in sperm DNA damage and abnormal mitochondrial membrane potential than normal patient group, and negative effects of varicocele induced oxidative stress were revealed.
