Abstract (EN)
In this thesis study, the antiproliferative effects of Aurora kinase inhibitor ZM447439 on Luminal A breast cancer and triple negative breast cancer cells were determined. For this purpose, MCF-7 was used as a Luminal A breast cancer model and MDA-MB-231 cell line was used as a triple negative breast cancer model. In the experiments, cell viability, cell index values obtained from xCELLigence real time cell analysis system, mitotic index, BrdU cell proliferation analysis and apoptotic index were evaluated as cell kinetics parameters. In the experiments, ZM447439 concentrations of 20 μM, 40 μM and 80 μM were applied for both cell lines, and the concentration of 40 μM ZM447439 was determined as the IC50 dose. When the data obtained as a result of all applications were examined, a significant decrease was observed in cell viability, cell index, mitotic index and BrdU % values in both cell lines, while a significant increase was observed in apoptotic index values.
