The effects of resveratrol and ?-lipoic acid on some biochemical parameters in tissue and plasma of aged rats induced by 7,12-DMBA

Abstract (EN)

In the study, 16 months 42 male rats are used. Rats are randomly divided into groups and groups are determined as control, DMBA, DMBA+?-LA and DMBA+R. 55 mg/kg 7,12-DMBA, which is solved in sesame oil, intraperitoneal carried out rats except for control group. After the DMBA performing, 25 mg/kg ?-LA and 25 mg/kg resveratrol are injected intraperitonal into DMBA+?-LA and DMBA+R groups, respectively and antioxidant treatment is continued for six weeks. Vitamins ADEK, cholesterol, vitamin C, MDA and glutathione molecules are studied by using HPLC and analysis of fatty acid methyle esters are studied by using gas chromatography after tissue and sera samples are given.According to our results, ?-tocopherol amounts in the groups of disease and DMBA+antioxidant groups decreased compared to control group in sera, erytrocytes and liver (p<0.05, p<0.01, p<0.001)). While erytrocytes MDA level in DMBA group was increased (p<0.001), it was determined lower in antioxidants groups than control group (p<0.001, p<0.0001). Cholesterol levels in erytrocytes of DMBA and DMBA+R groups (p<0.001) and sera of DMBA+?-LA group (p<0.01) and also kidney tissues of DMBA, DMBA+?-LA (p<0.01) found to be higher. However, it is determined reducing cholesterol level of liver tissue in DMBA group (p<0.001).According to gas chromatography results, rations of palmitic acid (16:0) in erytrocytes, sera and liver tissues were increased compared to control group (p<0.05, p<0.01, p<0.001). Nevertheless it was determined palmitoleic acid (16:1 n?7, n?9) was decreased in kidneys, sera and liver tissues (p<0.001, p<0.05). The ratio of stearic acid (18:0) was increased in liver of DMBA (p<0.01), DMBA+?-LA (p<0.05) groups whereas it was decreased in erytrocytes DMBA+R group (p<0.05). Also the ratio of linoleic acid (18:2 n-6) was decreased in livers of DMBA and DMBA+R groups (p<0.05) and sera DMBA (p<0.01) group. Althought the ratio of arachidonic acid (20:4 n?6) was increased in kidney tissues of DMBA (p<0.05) group and sera DMBA+?-LA group (p<0.05) but it was decreased in sera DMBA group (p<0.01). In addition the ratio of docosahexaenoic acid (22.6 n?3) was increased in all of the tissues (p<0.05, p<0.01, p<0.001). Saturated fatty acids ratio was decreased in kidneys of DMBA group (p<0.05) and sera DMBA and DMBA+ ?-LA and DMBA+R groups (p<0.05, p<0.01) whereas it was increased in erytrocytes DMBA (p<0.05) and DMBA+?-LA (p<0.01) groups but it was not changed in liver tissue (p>0.05).Key Words: 7,12-DMBA, Resveratrol, ?-Lipoic Acid, ?-tocopherol, Retinol, Glutathione (GSH-GSSG), HPLC and GC, Aged Rats.
