Abstract (EN)

The objectives of this study are purification and biological, serological andmolecular detection of TSWV in tomatos and peppers growing in Adana and Mersinprovinces between 2005 and 2006.Collected samples were firstly tested with ELISA and infection ratio wasdetermined as 45.4%. TSWV positives were coded and used for mechanicalinoculation. The symptoms were observed on E8, D2 and M5 isolates inoculated testplants in 5-15 days however, T1 inoculated ones showed in 1-2 months after TSWVinoculation.Only one protein of TSWV was observed in SDS-PAGE and molecularweight of it was estimated as 27 kDa.Centrifugation method was succesfully used for semi purification of TSWV.Column chromatography and PEG precipitation methods were not be successed forTSWV purification.TSWV virions were purified and four bands were obtained in SDS-PAGEand one band in agarose gel electrophoresis.Total RNA extraction was done from infected test plants and used for RT-PCR method. DNA bands were observed in respected sizes after the PCR productswere tested in agarose gel electrophoresis.Key Words: TSWV, ELISA, Survey, Mechanical Inoculation, RT- PCR
