Abstract (EN)

Many recent studies have shown that most non-culturable bacterial species are widespread and dominant in their environmental niche. The analysis of genomic material obtained from the environment (environmental DNA (eDNA)) by metagenomic methods provides the possibility of studying microorganisms that cannot be cultivated. Akkaya Dam, one of the oldest dams in Turkey, was built for irrigation purposes. In this study, the microbial flora of Akkaya Dam Lake in Niğde province and has had eutrophication problems for years, was determined by metagenomic analysis. The microbial flora was followed throughout a year by sampling from the same location every month. In the thesis study, it was observed that the abundance of bacteria in Akkaya reservoir varies seasonally, especially in the summer months, the predominance of genera showing eutrophication and fecal contamination increased. In addition, a large proportion of chemolithotrophic bacterial phyla, ordo and families were found in the lake. This shows the importance of watersheds in biogeochemical cycles. Therefore, it is important to protect the Akkaya lake area from chemical and urban sewage pollution.
