Abstract (EN)

The basic properties required in wound dressings should balance the moisture level in the wound, have gas permeability, be a barrier to protect the wound from external factors, be biocompatible, biodegradable, antibacterial, accessible, and most importantly include extra properties that accelerate wound healing. The aim of this study is to produce antibacterial wound dressing materials which contain allantoin and accelerate wound healing and tissue regeneration due to allantoin release by electrospinning method. In this thesis, polyurethane containing trimethylolpropane ethoxylate having 3 different molecular weights was synthesized. The materials formed by adding polycaprolactone to synthesized polyurethanes were characterized by FTIR, AFM, TGA, DTA and DSC. 1%, 3%, 5% allantoin molecules and 1% gentamicin sulphate were added to the polymers which were characterized and converted into wound dressing by electrospinning method. Morphological, mechanical strength and flexibility, biodegradability, antibacterial, allantoin release level, in vitro biocompatibility and cell adhesion tests of wound dressings prepared after this stage were performed. As a result, the obtained TMPE-based dressings are thermostable up to approximately 300 and are easily processable. Fiber diameters of the wound dressings are approximately 544±180, 1328 ± 349, 1495 ± 492 nm. The structures are flexible, homogeneous and have mechanical properties of approximately 4,6 MPa. Each dressing material produced a regular release of allantoin for 72 hours and showed antibacterial properties against gram negative (-) and gram positive (+) bacteria. Their biocompatibility is around grade 1. As a result, with the targeted study, TMPE based polyurethane/polycaprolactone combination, potential wound dressings containing antibacterial and allantoin have been developed.
