Abstract (EN)

In this thesis study, it was aimed to investigate the mass production possibilities with different basic nutrient media and plant growth regulator combinations in seed materials taken from Uzun and Siirt pistachio varieties (P. vera L.) cultivated in İkizce village of Nizip district of Gaziantep province. Different basic nutrient media and plant growth regulators were applied in the study, the highest sterile and germinating embryo germination rate was obtained from 1.0 mg/l BAP + 0.1 mg/l IBA + 0.25 mg/l GA3 containing 71% in Siirt variety and 69% in Uzun variety. In tillering experiments; When MS and WPM basic nutrient media were compared, the highest average shoot number was 2.6, and shoot length was 3.2 cm in Siirt variety and MS nutrient medium containing 1 mg/l BAP, 0.1 mg/l IBA and 0.25 mg/l GA3. In tillering experiments with different BAP concentrations, the highest average shoot number per explant was 2.6 in both varieties. The average shoot length was 2.6 cm in the Siirt variety and 2.2 cm in the Uzun variety. Siirt variety was higher than the Uzun variety, but this difference was not statistically significant and the highest results were obtained in MS nutrient medium containing 1.0 mg/l BAP + 0.1 mg/l IBA + 0.25 mg/l GA3. Shoot length, root number, and root length varied according to the treatments. The medium which provided the best rooting was obtained from ½ MS nutrient medium containing 1 mg/l IBA + 1 mg/l IAA + 1 mg/l NAA and 1 g/l activated carbon and in these media, the average root length per explant was determined as 2.53 cm in Siirt variety and 2.67 cm in Uzun variety. When the varieties in question were compared in terms of root number, the average root number in the Siirt variety was 3.20, and in the Uzun variety; It was determined to be 2.80.
