Abstract (EN)

This study was conducted to investigate the effect of adding Apilarnil to semen at different doses in vitro on the cryopreservation of semen. In the study, semen taken from 6 adult male New Zealand rabbits was diluted with Tris + egg yolk. Different doses of Apilarnil (0,5%, 1%, 1,5% and 2% doses) were added to the diluted semen and frozen in liquid nitrogen vapor. In this study, It was determined that 0,5% and 1% doses of Apilarnil positively affected total and progressive motility, 1% dose positively affected medium fast motility and 0,5% dose affected rapid motility, while 2% Apilarnil dose negatively affected motility parameters. A decrease in the VCL value was calculated at the 1,5% and 2% doses, in the VAP value at the 2% dose, and in the ALH value (p<0.001) at the 1,5% and 2% doses. of apilarnil. There was an increase in LIN value at the 1,5% dose and in the WOB value (p<0.001) at the 1%, 1,5% and 2% doses of Apilarnil. Moreover, after equilibration, It was determined that there was an increase in ALH value (p<0.001) at the 1%, 1,5% and 2% doses of Apilarnil. It was determined that a 1% dose of Apilarnil increased the HOST value, decreased the total and head-attached abnormal spermatozoon rates, increased the live spermatozoon rate and decreased the dead spermatozoon rate. It was determined that all doses of apilarnil reduced the rate of acrosomal damage. It was determined that the 1% dose of apilarnil increased high mitochondrial membrane potential (YMMP) and decreased low mitochondrial membrane potential (DMMP) (p<0.01). It was determined that the 2% dose of apilarnil decreased YMMP and increased DMMP (p<0.01). It was determined that the 2% dose of apilarnil increased MDA, the 1,5% and 2% doses decreased the GSH-Px value, and the 2% dose decreased the CAT value (p<0.01). As a result, it has been suggested that adding a 1% dose of Apilarnil to the semen extender in the cryopreservation of rabbit semen will be beneficial and contribute to the successful freezing of the semen.
