Determination of anticholesterol activities and their resistance to the artificial gastric juice and intestinal juice of Enterococcus bacteria, which are isolated from the fresh white cheeses

Abstract (EN)

In this study, a total of 8 Enterococcus strains including Enterococcus faecium DSMZ 20447 and Enterococcus faecalis NCDO 581 as standard strains were tested for their survival in artificial gastric and in intestinal fluid juice. Also, the effect of different bile salt concentrations and anti-cholesterol activities of the strains were estimated. Spectrophotometric method were used for the studying antioxidant activities of the strains by measuring the percentage of reduction.Enterococci were subjected to artificial gastric juice on different pH?s and on different periods. It was observed that all the strains survived in all pH?s at the beginning. Also, it was observed that only Enterococcus faecium RT 81 and Enterococcus faecalis NCDO 581 strains survived at pH 2,0 at the 60th minute. At the same minute, in pH 3,0; it was observed that other than Enterococcus faecium DSMZ 20447 standard strain, all the strains showed decreasing density of survival. In pH 4,0 ; it was determined that all the strains survive. At 180th minute, in pH 2,0 ; none of the strains showed any sign of survival. However, at the same minute, in pH 3,0 ; it was observed survival at Enterococcus faecium RI 41 strain and at Enterococcus faecalis NCDO 581 standard strain. Again at the same minute, in pH 4,0 ; it was observed that all the strains showed decreasing density of survival.It was determined that all the strains that are tested could survive for 240 minutes in the artificial intestinal fluid. It was determined that while Enterococcus faecium RT 81 strain has the highest unit of survival, Enterococcus faecium DSMZ 20447 standard strain has the lowest unit of survival. It was determined that, Enterococcus faecalis NCDO 581 standard strain was resilient to both artificial gastric juice and intestinal fluid.The cholesterol reduction in Enterococcus strains were determined at 0,0%; 0,6%; 0,15% and 0,30% percentages in bile containing environments. In nutrition environment containing 30% of bile, it was determined that Enterococcus faecalis RT 122 strain has the highest unit of cholesterol reduction. While Enterococcus faecalis RT 122 strain performed 81% of cholesterol reduction, it was determined that with 4% of cholesterol reduction, Enterococcus faecalis RI 6 strain has the lowest unit of reduction in non-bile containing nutrition environment.Antioxidant activities of the strains are researched. While, Enterococcus faecalis NCDO 581 standard strain showed the highest percentage of antioxidant activity, it was determined that Enterococcus faecalis RT 122 strain showed the least percentage of antioxidant activity.
