Effects of resveratrol on oxidative and histopathological changes with SIRT1 gene expression in brain neuroinflammation model

Abstract (EN)
Aim: The aim of this study is investigating the effects of resveratrol and EX-527 on LPS-induced neuroinflammation rat model brain as molecular genetic, biochemical, cognitive and histopathological and whether there is any neuroprotective activity. Material and Method: This study was carried out with 55 Wistar albino male rats. SIRT1 gene expression, biochemichal analyses and histopathological examination were made in rat brain tissue. Analysis of cognitive effects of disease model was practiced by Morris water maze. Results: In this study, MDA levels of brain tissues in Res and Res+LPS groups were found statistically higher than K, LPS and EX-527+LPS groups (p<0.05). SOD activities in brain tissues of EX-527+LPS were increased in comparision with K and LPS groups statistically (p<0.05) while decreased in Res and Res+LPS groups was significantly compared to K group (p<0.05). Expression of SIRT1 brain gene in EX-527+LPS group was relevantly lower than other groups (p<0.05). There was a considerable rise in SIRT1 gene expression of Res+LPS group in comparison with K group (p<0.05). Histopathologically, brain sections of K and Res groups had normal morphological characterictics and neurons in LPS group showed degenerative alterations as there were a few degenaration in Res+LPS groups. Degenerative neuron density in Res+LPS group decreased compared to LPS group prominently and neuron degeneration was more elevated in EX-527+LPS group than K and Res groups (p<0.05), and had more less neuron degeneration density than LPS group (p<0.05). Conclusion: It has been concluded that LPS-induced neurodegeration can be prevented by enhancing SIRT1 gene expression with Resveratrol.
