Abstract (EN)

Aim: Studies have shown that RML, which has been reported to have antioxidant/anti-inflammatory effects, should be evaluated as a new neuroprotective agent in the treatment of DN. To this end, DN models were created and the effect of RML was comprehensively investigated. Material and Method: In the in vitro phase, DRG cells were exposed to high glucose, RML was administered, and cell viability was examined using the MTT method. In the in vivo phase, 40 male Balb-c mice were divided into four groups: Control, DN, DN+RML-3, and DN+RML-6 (n=10). STZ (150 mg/kg) was injected into the groups other than the Control group. Animals with BG levels ≤ 250 mg/dL 72 hours later were considered diabetic. A 21-day period was observed from the onset of diabetes. Subsequently, while no treatment was administered to the control group, intraperitoneal administration was performed for 15 days to the RML groups (3-6 mg/kg RML). Results: The findings indicate that RML application protected DRG cells (p<0.05). Furthermore, RML reduced BG levels compared to the DN group, increased the pain threshold, improved oxidative stress and inflammation parameters (p<0.05), and inhibited apoptosis by repairing histological damage (p<0.0001). Conclusion: In conclusion indicate that RML may be a target molecule for preventing DN progression and complications. Key Words: Diabetes, DRG, Neuropathic Pain, Ramelteon
