Abstract (EN)
In this study, new amperometric enzyme electrode for determination of glucose oxidase was developed. Glucose oxidase enzyme has been immobilized at media prepared with paratoluensulphonate of pyrrole in polypyrrole-paratoluensulphonate film crosslinking via glutaraldehyde method. For this reason; optimum conditions of Pt/ polypyrrole-paratoluensulphonate electrode were determined. Glucose detection is based on the oxidation of hydrogen peroxide at 0,30 V produced by the enzymatic reaction on the enzyme electrode surface. The effects of pH and temperature on the response of the glucose enzyme electrode were investigated. The immobilized of glucose oxidase KM(app.) and Vmax(app.) values were found. Determination of glucose was carried out in biological fluid (in blood) by enzyme electrode. Interference effects were investigated on the amperometric response of the enzyme electrode .Key Words :Glucose, glucose oxidase, enzyme electrode, biological fluid, polypyrrole, paratoluensulphonate
