Ill IN VITRO PROPAGATION OF Centaurea tchihatcheffii (M.Sc. Thesis) Çiğdem Alev ÖZEL GAZI UNIVERSITY INSTITUTE OF SCIENCE AND TECHNOLOGY September 2002 ABSTRACT The research was carried out to study endemic and commercially important ornamental Centaurea tchihatchiffii. High frequency of contamination was noted down, when surface sterilizing the seeds. Least uncontaminated seeds were obtained from 50% commercial bleach with application time of 10 minutes and 60% commercial bleach with application time of 10, 20, or 30 minutes. Only one seed out of 250 seeds placed at +4°C for 13 days under dark, germinated after 150 days. The green material from wild was double sterilised using 30% commercial bleach for 5 minutes; however, it resulted in partial success of 33.3%. However, after 5-6 weeks in culture, the contamination reappeared from the systemic fungus or bacteria. The best micropropagation results were obtained from MS medium containing 0.5 mg/l BAP and 0.01 mg/l CPA. Developing shoots were limited, dwarf and dominantly vitrified. Eighty percent sterilisation was achieved from immature embryos. The best shoot regeneration was achieved on MS medium containing 1 mg/l Kinetin - 0.25 mg/l IBA. Rooting study was carried out both under in vitro and ex vitro conditions. The best rooting response was obtained from shoots treated with 500 ppm IBA for 10 minutes under ex vitro conditions after 10 days. Under in vitro conditions, rooting started after 12 days on MS medium containing 0.25 mg/l IBA. The rooting medium containing 0.5 mg/l IBA resulted inIV development of multiple shoots that was very similar to micropropagation.
