Effect of Centaurea urvillei on toxoplasma gondii

Abstract (EN)
In our study; It was aimed to ensure the reproduction of T.gondii parasite, which is produced by ex vivo culture method in Turkey, without losing its virulence properties by ex vivo cultivation method in Vero cells, and to investigate the antiparasitic drug activity of Centaurea urvillei plant on this parasite. Centaurea urvillei plant was collected from Afyon province, dried and extracts were prepared by taking the parts that can be used for extraction and dissolving them in methanol, n-hexane and chloroform solutions. Vero and L929 cells were then thawed for passage. Cell count was performed by staining with trypan blue. On the other hand, Toxoplasma gondii was removed from the liquid nitrogen tank and thawed. Parasite count was done by staining with trypan blue. In vitro cytotoxicity evaluation was performed on L929 cells of Centaurea urvillei plant. The CC50 values of n-hexane, methanol, chloroform and pyrimethamine were determined as 110.4 µg/ml, 99.6 µg/ml, 157.7 µg/ml and 189.8 µg/ml, respectively. As a result, it was determined that Centaurea urvillei plant did not show cytotoxic activity. Then, ex-vivo cultivation of T.gondii was carried out. T.gondii isolate was cultivated in Vero cell and left for 72 incubations. At the end of 72 hours, infection occurred and the evaluation of the C.urvillei plant at different concentrations was made to look at the effect on T.gondii. The infected Vero cells were incubated for 48 hours after the addition of the control drug and the extract. Afterwards, it was stained with Giemsa and the number of tachyzoites in each well was determined with a light microscope, and the % anti-tachyzoite (inhibition) activity was calculated. After determining the IC50 value, the selectivity index (SI) was found to determine whether it has antiparasitic activity and it was found that Centaurea urvillei plant showed antiparasitic activity on T.gondii, and it was found that it was statistically significant, and its effect on T.gondii can be seen as a step in future drug studies.
