Investigation of cytotoxic/apoptotic and antimicrobialeffects of some benzimidazol metal complex (Zn, Co)compounds on various cancer cells

Abstract (EN)
Cancer disease has become the biggest health problem that affects human life both spiritually and financially. The work done to solve this problem is also ahead of the studies in the field of health today. One of the most predictable solutions for the treatment of this disease today is to develop effective drugs against this disease. For this purpose, metalcontaining benzimidazole compounds have been seen among the most suitable candidates in recent years. In this study, the cytotoxiceffects of nine benzimidazole-derived compounds containing cobalt and zinc on A549 (non-small cell lung carcinoma), BEAS-2B (healthy lung epithelial cell line), HCT116 (colorectal carcinoma) and Hep3B (hepatocellular carcinoma) cells has been examined using Trypan blue, MTT, Caspase-3 and Western Blotting methods. The cytotoxic effect of all compounds was compared with cisplatin used as reference compound. Their antimicrobial effects were also determined by the Minimum Inhibition Concentration (MIC) method. Two of the compounds used in the study (Compounds 4 and 9) were found to have high cytotoxic effects on Hep3B and HCT116 cells. Compounds 6 and 8 showed a high cytotoxic effect on A549 cells, like cisplatin, but a lower cytotoxic effect on BEAS-2B cells than cisplatin. MTT analysis showed that the IC50 values of cisplatin, Compound 6 and Compound 8 on A549 cells at 72 h were 4.31, 6.27 and 4.66 μg / mL, respectively. On the other hand, their IC50 values on BEAS-2B cells at 72 h were determined as 1.78, 54.88 and 34.34 μg/mL, respectively. The compounds have also been found to have a high antimicrobial effect on bacteria and yeasts.
