Membrane filtration and conventional methods in microbiological quality analysis of various water samples and determination of antibiotic resistance

Abstract (EN)

In this study, two hundred and fifty water samples (tap water, spring, well and spa water) were collected between 13.08.2008 and 29.01.2010 in Ankara. 250 water samples were analysed for total mesophilic aerobic bacteria, total coliform, Escherichia coli, Enterococcus spp., Staphylococcus spp. and Gram negative fermentative and non-fermentative bacteria the membrane filtration and conventional methods.Of the 289 isolated strains, 153 were (52,9%) determined as coliform bacteria, 26 (9%) as non-fermentative and fermentative microorganisms, 34 (11,7%) as Enterococcus faecium, 26 (9%) as Enterococcus faecalis, and 35 (12,1%) as coagulase negative Staphylococcus (CNS), 15 (5,2%) as Staphylococcus aureus according to Bergey?s Manuel of Systematic Bacteriology. Also the antimicrobial susceptibility to various antimicrobial agent of 289 microorganisms isolated from different water samples were tested with Kirby-Bauer disc diffüsion method. All isolates were resistant to one or more antibiotics. Among (179) Gram negative bacteria isolates, the highest resistance was found for ampicillin, penicilin, ceftriaxone, carbenicillin (from 91,4 % to 100%). Among (110) Gram positive bacteria isolates, the highest resistance was also found for ampicillin, nalidixic acid, oxacillin, carbenicillin (from 70,6 % to 100%).According to our result the membrane filtration method is a simple and efficient alternative method than conventional method for the enumeration of bacteria from various water samples. As a result it is recommended that membrane filtration method is useful for enumaration of microbiological quality analysis of Drinking and Using Water Regulation. Besides all water samples have been high microbial contamination and showed potantial hazardous for public health.
