Abstract (EN)

The aim of this study was to investigate the clinical role of seminal fluid analyses, DNA fragmentation index (DFI) (by using aniline blue staining of histones), and level of seminal 8-OHdG. Patients (120) were divided into normozoospermia, asthenozoospermia and oligozoospermia according to seminal fluid analysis. This study investigated the differences of 8-OHdG levels and sperm DFI in these groups. The correlation of sperm DFI and levels of 8-HOdG was tested and the correlation with the seminal parametres were studied also. The result showed that 8-OHDG levels increased significantly (P= 0.004) with age, and reached its highest level among the participants aged ≥ 50 years. Sperm DFI showed significant differences among seminal fluid groups with p=0.003. Sperm DFI was negatively correlated with sperm concentration, motility and percent morphology (P= 0.001) while it was positively correlated with number immotile sperm (P= 0.001). Morover, there was a significant positive correlation between 8-OHGD and DFI (P= 0.001). This study concluded that testing DFI and seminal fluid analysis should be considered as complementary diagnostic tools in a comprehensive evaluation of male fertility
