Evaluation of effectiveness of cow colostrum on treatment and its effect on proteomic structure in blood serum in calves with cryptosporidiosis

Abstract (EN)

Aim of the presented study was investigate the effect of using colostrum combined with or without bicarbonate in addition to paramomycin, on the treatment outcome and serum proteomic profiles in calves with cryptosporidiosis. Inaddition, it was aimed to compare proteomic analysis results of high and low quality colostrums for the first time. Thirty Holstein calves younger than 21 days that were infected only with Cryptosporidium spp. were allocated into three equal groups. Paramomycin was administered 1x100 mg / kg for 5 days to all three groups. Additionally 250 ml colostrum was fed twice a day for 3 days to Groups 1 and 2. Additionally Group 1 recieved 6 g oral sodium bicarbonate twice a day before colostrum administration. Clinical examinations, fecal scoring and oocyst counts of calves were conducted before and 10 days following the initiation of treatment. Brix% and total protein (TP) levels were analysed from sera at day 0, 1, 2, 3, 5 and 7 days; immunoglobulin G (IgG), gamma-glutamyltransferase (GGT) measurements and proteomic analyzes were performed at day 0 and 3 day. Fecal scores decreased significantly (p <0,05) on the second day of the treatment in Group 1 and 2 comparing to pretreatment levels. But same decrement was not observed in Group 3 (control group) till the 8thday of the treatment. Similarly, oocycts counts decreased in Group 1 and Group 2 respectively on 2 ndand 3 rdday of the treatment however decrement was detected in Group 3 on the 10thday (p<0,05). Decrement of IgG, TP, Brix% and GGT showed that large mollecules were not absorbed enough in spite of colostrum treatment. Colostral treatment used in Group 1 and 2 changed the levels of A1AT, SAA, ACTG, HP20 proteins and A1BG, S39AB, CTHL1, ACTB APOA4 proteins respectively but there was no differences for protemic structure in Group 3. Results showed that using colostrum in the treatment effects the calf serum proteomic. Inaddition, 8 proteins in high quality and 11 proteins in low quality colostrum were detected high concentration. In conclusion, using paramomycin with bicarbonate and colostrum gave better outcome in cryptosporidiosis treatment.
