Abstract (EN)

In this study, anticholinergic, antidiabetic, antioxidant, antibacterial activities and phenolic content of the extract of Cyclotrichium longiflorum plant prepared with a solvent mixture consisting of methanol, ethanol, distilled water (72.5/14.8/12.7, v/v/v) were evaluated. In the study, 2.2-Diphenyl-1-picryhydrazyl (DPPH), 2.2 azinobis-(3-ethylbenzothiazoline-6- sulfonic acid (ABTS) and Iron reducing antioxidant power determination (FRAP) methods were used for antioxidant activity analyses. In addition, total phenolic and flavonoid content determination experiments were performed in the plant. Acetylcholinesterase (AChE), butyrylcholinesterase (BChE), alpha-amylase and alpha-glucosidase enzyme inhibition activity experiments were performed for anticholinergic and antidiabetic activities. Disk diffusion method and Minimum İnhibitory Concentration (MIC) were investigated against Escherichia coli and Stahylococcus aureus bacteria for antibacterial analysis. LC-MS/MS method was used for detailed analysis of phytochemical components of the plant. As a result of the analyses, IC50 values of antioxidant effect of C. longiflorum plant were calculated, and it was determined that the plant had high antioxidant effect as a result of three experiments. It was also seen. In the first of the antibacterial experiments, the disk diffusion method, the plant formed a good zone for both bacteria and the results were compared with positive and negative controls. Significant antibacterial activity was seen in MIC results. In the enzyme inhibition experiments, it was determined that the plant had a strong inhibitory activity against AChE and BChE enzymes and it was concluded that the plant had a significant anticholinergic effect. Rather than the expected inhibitory effects in α-amylase and αglucosidase enzymes, an opposite increase was observed in the activity of these enzymes. As a result of LC-MS analysis, it was determined that the extract contained significant flavonoid compounds such as naringin and hesperidin. It has been seen in the literature that these compounds bind to the active sites of enzymes and have an activator effect. Similar results were obtained in this study. In addition to naringin and hesperidin, rutin hydrate, benzoic acid and eridictyol were found in C. longiflorum plant in the LC-MS/MS method.
