Abstract (EN)
Hypoxic ischemic encephalopathy (HIE) of prenatal, natal and postnatal factors It is a brain injury that occurs as a result of systemic hypoxia and decreases cerebral blood flow. Despite all the developments in medicine, mortality and morbidity rates due to HIE are still high. Currently, only hypothermia is available as a supportive therapy in HIE. Our aim in this study is to show the effect of ascorbic acid and erythropoietin in HIE treatment. Wistar rats were used for this purpose. The experimental animals were divided into 8 groups with 7 rats in each group. The rats in the HI group were exposed to hypoxia by carotid ligation and administration of 8% oxygen for 2 hours. Ascorbic acid (1000mg / kg, 2000mg / kg, 5000mg / kg), erythropoietin (2000 U / kg, 5000 U / kg), ascorbic acid + erythropoietin (5000 U / kg + 2000mg / kg) by intraperitoneal injection ) and saline was given. At the end of the experiment, the rats were decapitated. Subsequently, the brain tissues of the rats were rapidly removed and stained using TUNEL techniques. Caspase 3, Caspase 8, Caspase 9, Malondialdehyde, Total Antioxidant Capacity, Total Oxidant Capacity, Hypoxiainducible factor 1 alpha activity were studied by spectrophotometric method. In our study, it was determined that 5000mg / kg vitamin C was given to the group, which reduced HIF-1α and apoptosis compared to the control group. Caspase activity was found to be lower in the 5000mg / kg vitamin C group compared to the erythropoietin group. In our study, when delayed neuronal damage was examined after hypoxic ischemic injury, ascorbic acid and erythropoietin were thought to decrease hypoxiainducible factor 1 alpha caspase activity and apoptosis as a new hope.
