The efficacy of cyclosporine-a in the experimental sciatic nerve injection injury

Abstract (EN)

In this experimental study the efficiacy of cyclosporin A in peripheric nerve injection injury by Penicillin-G was investigated with electrophysiological and light microscopical investigation. 20 male albino rats were used ( weighted 250-300 gr) in this study. Under general anaesthesia, right sciatic nerve was explored in each animal . Normal physiologic compound muscle action potantials were recorded before injection injury. Then injury was made by intraneural Penicillin-G injection. After injury compound muscle action potantials were recorded again. The animals were divided in 4 groups. Loading dose of cyclosporin A of 20 mg/kg was given intraperitoneally to the first group at 30th minute after injury, and then it continued with the dosage of 10 mg/kg once in a day for 3 days with the same route. In other two groups cyclosporine-A was given at 8th and 24th hours after injury with the same dosage and route just as in group I. Control group did not take any medication except prophylactic antibiotics. 4 weeks after injury , compound muscle action potantials were recorded in all the groups and then animals were sacrified. The tissue samples were taken from each animal , light microscopic investigation was performed. After treatment improvement of the compound muscle action potantial amplitude of the group I was found statistically significant (p < 0.05). In the other groups the improvement was not significant statistically ( p > 0.05). Both electrophysiological and histopathological investigations demonstrated the neuroprotective effectiveness of cyclosporin A if it was given in the first 30 minutes after injury. But in delayed period ( 8 and 24 hours ) cyclosporine A therapy does not have significant recovery statistically.
