Abstract (EN)

In this study, the β-mannanase enzymes derived from Bacillus sp. strains which is isolated from different environments, were produced and characterized. Wide range of pH value and temperature were tested on bacteria to produce and observe reproductive capabilities on solid plate. Different chemicals, inhibitors, schelators, detergent and metal ions effect on enzyme activities was observed. SDS-PAGE analysis made for MAN23 enzyme to determinate molecular weight which is estimated in between 40kDa, 15kDa and were formed in two bands. Optimum enzyme activity was observed at 60°C, pH 6. The enzyme was retained its activity in between 30°C and 80°C temperatures around %93. Enzyme showed that mesothermophill property. Enzyme presented acidic-neutral stability for pH range of 6-8 with retained its activity around %52. MnCl2 and β-merkaptoethanol induced enzyme activity %117 and %108 respectively. The enzyme has shown tolerance for only %3, %5 and %10 NaCl concentration retained around %52. Also washing tests were showed that MAN23 enzyme has managed to clear %100 of baby food from fabric. TLC analysis showed that enzyme was hidrolized mannan to D-mannose, mannobios and mannotriose. According to these results MAN23 enzyme shows thermotolerant, thermophile, acidophile, halotolerant and acidostable characters. Due to these properties MAN23 β-mannase enzyme can be used in detergent in, food and feed industry and recycling of biological waste.
