Detection and characterization of virus diseases and phytoplasma in rose growing in the Eastern Mediterranean region

Abstract (EN)
This research was carried out between 2020-2021 in order to determine and characterize the presence of virus and phytoplasma diseases that cause harm to roses grown in decked and open landscape and recreation areas in the Eastern Mediterranean Region of Adana and Mersin provinces symptomologically and molecularly. During the surveys carried out, symptoms such as mosaic, chlorosis, deformation, necrotic spots and stunting were observed on the leaves of rose plants. Rose samples showing signs of a virus or virus like disease were collected from the region, all of these samples were tested by RT Polymerase Chain Reaction (PCR) with different primers specific for the coat protein gene of ArMV, ApMV, PNRSV, SLRSV, RRV, RSDaV, RoLCV viruses. However, according to the test results, the tested plants were found to be negative for the viruses in question. PCR products obtained from direct and nested PCR results for suspicious samples showing signs of phytoplasma from recreation areas in Seyhan and Çukurova districts were used to perform RFLP studies using the EcoRI enzyme. In the samples taken, the presence of a disease caused by phytoplasma was revealed both symptomologically and molecularly. As a result of restriction the pathogen giving 1250bp band with the EcoR1 enzyme in Nested PCR, bands of 750 and 500 bp were determined and it was confirmed that it was a phytoplasma.
