Effect of different nutrient media and irradiation doses on haploid embriyo and plant formation in Galia and Kırkağaç melon (Cucumis melo L.) genotypes

Abstract (EN)

Use of haploid plants in shortening the breeding period and obtaining 100% homozygote plants in short time are important tools in plant breeding. In this research, the effects of combinations of three different media and three different pollen irradiation doses on embryo and haploid plant obtention in Galia and Kırkağaç melon types were investigated. In the study K-7, K-12 and K-13 genetypes from Galia type; K-17 genetype from Kırkagac type of Kuzey Agripark Company were used and this research was carried out Kuzey Agripark Company Greenhouses in 2015-2016 years. Comparasion of the melon types in terms of the average seed number, Galia types gave better results than the Kırkağaç type. Besides, 350 Gy pollen irradiation doses decreased the number of seeds obtained. The best irradiation dose was found as 300 Gy in terms of average seeds number for both melon types. Likewise 300 Gy dose gave better results on embryo and plant growing from embryo. According to the results, obtaining haploid embryo and conversion of embryo to plant depend on irradiation dose and medium. The highest percentage of embryos and plant were found in Galia type melon irradiated with 300 Gy. Therefore, the use of 300 Gy irradiation dose and MS nutrient media for obtaining haploid plant is recommended for further studies.
