ABSTRACT The Investıgatıon Of Potentıal Protectıve Effects Of Ellagıc Acıd Agaınst Reproductıve Damage Induced By Irınotecan In Male Rats Aim: Irinotecan is a camptothecin derivative antineoplastic and immunosuppressive drug. Ellagic acid is a powerful anticarcinogenic and antimutagenic natural phenolic compound. This study was planned to demonstrate the protective effects of Ellagic acid against reproductive damage caused by Irinotecan. Material and Method: In our study 40 Sprauge-Dawley rats were used (n=10). Groups: Control+CMC, IR, IR+EA, EA At the end of the study testis samples were processed by routine tissue techniques, embedded in paraffin. 5μm thick sections of tissues were cut, stained with H&E and caspase-3 examined under a Leica DFC280 light microscope by Leica Qwin and Image Analysis System. Results: In control and EA groups, testis showed a normal histological appearance with H-E procedure. In IR group, testis tissue showed vascular congestion, oedema and vacuolisation in interstitial area, reduction in germ line cells in seminiferous tubules, squamotion in lümen, arrested spermatocytes in different stage in division were observed. These fındings were significantly decreased in IR+EA group. In biochemical analysis, IR lead to a significant increased in TBARS levels and significant decrease in GSH, SOD, GPx and CAT levels in testis tissue compared with other groups. In the IR+EA group there was an attenuated increased in TBARS levels and an increase in GSH, SOD, GPx and CAT activities. Conclusion: As a result of EA have protective effects againts the reproductive damage which created with IR. Keywords: Irinotecan, Testis, Ellagic Acid, Rat, Caspase-3.
