Investigation of the effects of asprosin application on feed, water consumption, weight gain and hormones effective in energy metabolism in male rats after weaning

Abstract (EN)

Asprosin is a new adipokine with 140 amino acids, which is thought to be effective in energy metabolism, mostly secreted from white adipose tissue. In studies on asprosin in the literature, the pathologically high serum asprosin concentrations of obese humans and animals revealed that asprosin is associated with obesity. Although most of the studies on asprosin have focused on obesity, there are studies showing that asprosin may play a role in the pathogenesis of many diseases such as diabetes and insulin resistance, metabolic syndrome, and PCOS. The effectiveness of asprosin in energy metabolism and its association with obesity suggest that asprosin may have an effect on appetite and weight gain. Considering these data, our study included healthy male rats administered asprosin daily starting from the 21st day of weaning; It was aimed to reveal the possible effects of asprosin administration on weight change, feed-water consumption and hormones that have an active role in energy metabolism in rats by daily monitoring of eating-drinking behaviors and weight changes. For this purpose, 24 male Sprague-Dawley rats weighing 35±2 grams on the 21st day of weaning were used in our study. The animals were divided into two main groups (n=12) as control and asprosin, and asprosin was administered intraperitoneally (500 ng/kg) to the asprosin group between 13:00 and 15:00 every day. As in the asprosin group, intraperitoneal (1 ml/kg) saline was applied to the control group at the same times every day. Feed, water consumption and weight changes of the animals in both groups were monitored every day at 13:00 and recorded. At the end of the study, asprosin, leptin, ghrelin, insulin, TSH, GH and corticosterone levels were examined from the blood samples taken from the decapitated animals to be studied with the ELISA kit. Biochemically, lipid profile, LDL, HDL and TG levels were examined. In line with the data obtained; Compared to the control group, asprosin administration increased the feed and water consumption and weight of healthy male rats, but there was no statistical difference. When the serum hormone levels were examined, it was observed that there was a significant increase in corticosterone levels in the asprosin group compared to the control group. When analyzed biochemically, LDL levels were found to be statistically lower in the asprosin group.As a result, it was determined that the administration of asprosin in healthy male rats did not have a significant effect on food intake and weight change, and there was a significant increase in corticosterone levels in the group that received asprosin. As a result, it was determined that the administration of asprosin did not have a significant effect on food intake and weight change in healthy male rats, and there was a significant increase in corticosterone levels in the group that received asprosin.
