Abstract (EN)

In this thesis study, it was aimed to identify Calpastatin (CAST) and Diacylglycerol acyltransferase1 (DGAT1) gene polymorphisms in Central Anatolian Merino, Pırlak, Romanov and Suffolk sheep breeds raised in Eskişehir Province. Blood samples taken randomly from a total of 176 sheep (Central Anatolian Merino (137), Pırlak (8), Romanov (10) and Suffolk (21)) were used as material in the study. PCR-RFLP method was used to determine CAST and DGAT1 genotypes. DNA isolation from the studied samples was performed using a DNA isolation kit. PCR products amplified using appropriate primers were treated with MspI and AluI restriction enzymes, respectively. CAST and DGAT1 genotypes were determined using the band patterns obtained after electrophoresis in 2% agarose gels. In terms of CAST gene, two different alleles, M and N, were studied and the frequencies of M and N alleles were calculated as 0.67-0.33, 0.75-0.25, 0.75-0.25 and 0.76-0.24 in Central Anatolian Merino, Pırlak, Romanov and Suffolk sheep breeds, respectively. In terms of CAST gene, it was found that Pırlak sheep breed was in Hardy-Weinberg genetic equilibrium, while Central Anatolian Merino, Romanov and Suffolk sheep breeds were not in Hardy-Weinberg genetic equilibrium (P<0.05). Two different alleles, C and T, were studied for DGAT1 gene and the frequencies of C and T alleles were calculated as 0.60-0.40, 0.25-0.75, 0.80-0.20 and 0.31-0.69 in Central Anatolian Merino, Pırlak, Romanov and Suffolk sheep breeds, respectively. In terms of DGAT1 gene, it was found that Central Anatolian Merino, Romanov and Suffolk sheep breeds were in Hardy-Weinberg genetic equilibrium, while Pırlak sheep breed was not in Hardy-Weinberg genetic equilibrium (P<0.05).
