Abstract (EN)

Drosophila melanogaster is one of the most preferred model organisms due to its advantages such as being able to continue its culture in the laboratory condition, adapting to different ecological conditions and producing a large number of offspring in short time. It is important to culture model organisms using artificial diets in the laboratory conditions and to ensure the continuity of these cultures. Previous studies show that antibacterial, antifungal and anthelmintic substances added to artificial diet media are used to prevent microbial contamination and to obtain healthy individuals under laboratory conditions. Griseofulvin, an antifungal agent used in this study, was added to the artificial diet of D. melanogaster at concentrations of 50, 100, 200, 400 and 800 mg/L to investigate the effects of larvae, pupae and adult stages on survival rate and development time of the insect. Additionally, the same griseofulvin concentrations were exposed to D. melanogaster adult individuals. As a result of this, the oxidative stress markers, the amount of protein carbonyl (PCO), the amount of malondialdehyde (MDA) and the activity of glutathione-S-transferase (GST), which an enzyme involved in detoxification, were measured in adults. All concentrations resulted in reduced survival rate in all larval, pup and adult stages compared to the control; 200, 400 and 800 mg/L griseofulvin concentrations are resulted in a prolonged development time.The amount of MDA was significantly increased at the highest concentrations of griseofulvin, 400 mg/L and 800 mg/L, compared to the control and GST activity was shown to increase at these concentrations. It was shown that the amount of PCO increased at 50, 100 and 200 mg/L griseofulvin concentrations compared to the control. According to these results, it was observed that griseofulvin had a negative effect on the insect, especially at the highest concentrations tested, 200, 400 and 800 mg/L.
