Prof. Dr. Galip KAŞKAVALCIIn this study, segregating F2 populations were obtained from crosses of M. arenaria race 1,race 2, M. incognita, M. hapla resistant recombinat inbred lines Lucia, Shelby and Manteo withsusceptible lines Batem1 and Batem2. F2 indivuduals were phenotyped with M. javanica race 1 in atest including parents and F1 plants of the corresponding crosses. Resistance segregated 1:3 in F2populations, indicating the presence of an recessive resistance gene.In order to determine molecular markers linked to mj gene, 37 RAPD, 14 SCAR, 23 SSR,100 AFLP (EcoRI-MSeI) and 112 SRAP were used to screen the parents for polymorphism. 26 of the37 RAPD resulted with amplification product but none were polymorphic. Smilarity, 7 of the 14SCAR gave bands but none were polymorphic. 23 SSR primers with known map locations were usedto scren parents. Only one gave polymorphism but it was not linked to mj gene. Of the 100 AFLPprimers, 92 produced bands and two yielded candidate markers (E-ATT/M-CAA ve E-AAC/M-CTG)linked to mj gene. These two bands were cut of polyacrylimide gel, cloned and sequence analysed.Primers designed from the sequences did not yield polymorphic bands between parents. In addition,the sequences did not contain any restriction site which could have been used to convert them toCAPS markers.The two sequences obtained from AFLP polymorphic marker were used premiarly to designD1F, D1R, D17F and D17R primers. SRAP forward and reverse primers were used in combinationwith these four spesific primers to search for polymorphism between parents. Of the 112 primercombination 11 yielded polymorphism between parents and F2 individuals were screened with thesemarkers. MapMaker Exp 3.0 software was used to analyze the 11 markers.
