Synthesis, characterization and investigation of some biological activity of new hydroxamic acids and metal complexes containing active pharmaceutical ingredient

Abstract (EN)
In this work, paracetamolacetohydroxamic acid and potassium barbitalacetohydroxamate and their transition metal complexes were synthesized starting from the drug active ingredients. Spectroscopic, magnetic and thermal properties and some biological activities of the compounds were investigated. Synthesis section consists of two stages. In the first stage, synthesized acetoethyl esters of paracetamol and barbital were converted to paracetamolacetohydroxamic acid and potassium barbitalacetohydroxamate. In the second stage, metal complexes were prepared by reaction of ligands with metal salts in the aqueous media. The structures of ligands and complexes were characterized by elemental analysis, IR, 1H-NMR, and Mass spectroscopy and X-ray diffraction techniques. Magnetic susceptibilities of complexes were measured and spin-only magnetic moments were calculated. As a result of the studies, metal/ligand ratio were found 1:1 for the paracetamolacetohydroxamate complexes and 1:1 for Mn, Co, Cd and 2:1 for Ni, Cu, Zn complexes of the barbitalacetohydroxamate and all complexes contain different number of water molecules. Inhibitory effect of synthesized compounds against urease and pancreatic lipase enzymes were analyzed. Paracetamolacetohydroxamic acid caused high and potassium barbitalacetohydroxamate caused intermediate urease inhibition. Co(II), Ni(II) and Zn(II) complexes of paracetamolacetohydroxamic acid and Co(II), Zn(II) and Cd(II) complexes of barbitalacetohydroxamate caused high lipase inhibition.
