Abstract (EN)
Janus kinase- signal transducers and activators of transcription ( JAK-STAT )signaling pathway, has been implicated in a variety of cellular functions, which is foundto be activated by more than 50 cytokines or growth factors JAKs are receptor mediatedtirozinaz kinaz which activate STAT. The STAT family plays critical roles in antiviraldefense, acue phase response, hepatic injury, repair, inflammation, transformation andhepatitis. In the liver, interleukin-6 stimulates hepatocytes to produce a variety of acutephaseproteins, also plays an important role in liver regeneration and protection againstliver injury. IL-6 induces mainly the activity of STAT3. IL-10 also plays a role in theactivaty of STAT3 in the liver.Total hepatectomy materials of the 45 cirrhotic patients undergone to livertransplantation ( LTx) due to chronic HBV or HCV hepatitis in Inonu UniversityTurgut Ozal Medical Center, Malatya are recruited in our study and sorted into 3 groups( n=15) according to the medication taken prior to LTx. Group A received Lamuvidine,Group B Adevovir and Group C received either interferon or interferon + ribavidin.STAT 1,2,3,5,5a,5b,Il6, IL10 are worked out immunohistocemically. The staining ofthe hepatocytes, bile duct epithelium, lymphocytes, macrophages in the portal triadkupffer cells, fibroblast in the stroma. The expression of STAT 2, 3 and IL6 in thehepatocytes are found to be significant in highest group. A compared to that of groups Band C. STAT2 is found to be statistically significant in group B compared to that ofgroup C but there was statistically significence of STAT 3 and IL6 expression betweenthese 2 groups.There was a reverse relation concerning the STAT 5a and STAT 2 expressionwhere as STAT5A expression was found to be atmost high in who take interferon groupC and least in who take Lamivudin group A. STAT1 and STAT 5 expression wereweak in all of the groups. STAT 5b expression was high all of the groups and there wasnot any significant difference of STAT 5b expression between in these groups. IL 10expression was lowest in group c and there was no difference between groupA and B.While, after a LTx some patients pose early rejection, some patients needminimum immunotherapy. Recurrence of the primary disease after LTx is anotherproblem. In the Liver STATs have many functions such as antiviral defence, liverregeneration, inhibition of the ischemi-reperfusion injury. STAT and its effect on ILexpression in patients, taking different medicationmay play a pivotal role in theimmunompdulation after LTx. These must be taken into consideration in the patientfollow-up.Key Words: Liver Transplantation, STATs, IL-6, IL-10, Lamivudin, Adefovir,Ribavirin, Inerferon
