Evaluation of changes by age in skeletal and heart muscle via using Western Blotting and immunohistochemistry

Abstract (EN)

Aging process begining with the birth contains all cellular changes which cause death of the organism in parallel with time. Determining decreasing or increasing molecules in particular tissues during aging and the effect mechanism of these molecules will contribute to our understanding of functioning of this process significantly. Because of this reason, our study aims at comparative examination of Type I Collagen, FGF-2, IGF-I and GDF-8 in the skeletal and cardiac muscle in the different age groups through the methods of immunohistochemical, SEM, TEM and Western blotting.In our study 5 groups were formed from prenatal and postnatal different age groups, 6 rats were put in each group. Groups were determined as the prenatal group (20th Day), neonatal group, 4-week group (prepubertal period), 6-month group (mature period) and 12-month group (begining of old-age period). Type I Collagen, FGF-2, IGF-I and GDF-8 immunoreactivities were identified in all groups. Western blotting analysis was evaluated for Type I Collagen in all groups in the skeletal and cardiac muscle. All findings are supported by research and analyzes of SEM and TEM.In our study, heart muscle as well as skeletal muscle, similar age related type-1 collagen increments were determined. On the other hand FGF-2 was intense in skeletal and heart muscle at newborn group. In 6th month group, IGF-1 immunoreactivity was not certain except sarcolemma and nucleus. GDF-8 expression was seen in all groups increased gradually until complete morphogenesis.Consequently, in the light of these findings, in our study in which important molecules in parallel with the aging were evaluated through the various methods as of the development of the skeletal and cardiac muscle it has been estimated that the skeletal muscle lives the aging process more effectively in comparison to the cardiac muscle, the cardiac muscle stabilize itself from the adult period to the old age period. These findings have been evaluated as result of the voluntary and irregular mobilization of the skeletal muscle in comparison to involuntary and regular working of the cardiac muscle.
