Simultaneous determination of paracetamol, pseudoephedrine, chlorpheniramine, dextromethorphan, 4-aminophenol and ephedrine in tablets by capillary electrophoresis

Abstract (EN)
A new capillary electrophoresis method was developed to analyse pharmaceutical preparations containing combination of paracetamol, pseudoephedrine, chlorpheniramine, dextromethorphan, 4-aminophenol and ephedrine simultaneously. Best result were obtained by use of 75 mM %15 ACN borat buffer as the background electrolyte. The separation was performed through a fused-silica capillary (75 µm internal diameter, 50 cm total length, 41 cm effective length) at 30°C with the application of 10 seconds of hydrodynamic injection at 50 mbar pressure and potential of 30 kV. Detection wavelength was 192 nm. Under these conditions, the migration times were found to be 10,885 min for paracetamol, 7,514 min for pseudoephedrine,6,523 min for chlorpheniramine, 5,845 min for dextromethorphan, 9,273 min for 4-aminophenol and 6,017 min for ephedrine. Linearity ranges for the method were 50- 900 µg/mL for PAR, 25- 120 µg/mL for PSO, 2- 10 µg/mL for KLO, 25-100 µg/mL for DEK, 5-20 µg/mL for 4-AF, 5-20 µg/mL for EFN. Limit of detection values were found as 12,38 µg/mL for PAR, 5,14 µg/mL for PSO, 0,41 µg/mL for KLO, 4,47 µg/mL for DEK, 0,97 µg/mL for 4-AF and 1,14 µg/mL for EFN. According to the validation study, it was proved that the developed method was accurate, precise, sensitive, specific and repeatable.
